      目的 探讨利奈唑胺诱导耐多药结核病患者周围神经病变的潜在机制。方法 收集临床耐多药结核患者的基本信息、不良反应等评估利奈唑胺对外周神经的影响;构建利奈唑胺长期给药SD大鼠模型考察坐骨神经的病理性改变。使用施万细胞模拟利奈唑胺暴露实验分析周围神经损伤与自噬的关系。结果 临床数据显示,有36.4%的患者使用利奈唑胺后出现周围神经相关症状。H E染色显示,利奈唑胺导致坐骨神经组织排列紊乱、间隙增大。WB结果表明利奈唑胺诱导的周围神经毒性与自噬相关。结论 长期使用利奈唑胺诱导的周围神经病变,其作用机制可能与激活施万细胞的自噬有关。
英文摘要:
      OBJECTIVE We aimed to explore the underlying mechanism of linezolid-induced peripheral neuropathy in MDR-TB. METHODS The basic information and adverse reactions of clinical MDR-TB patients were collected to evaluate the effects of linezolid on peripheral nerves. The pathological changes of sciatic nerve were investigated in SD rat model with long-term administration of linezolid. The relationship between peripheral nerve injury and autophagy was analyzed using Schwann cell simulated linezolid exposure assay. RESULTS Clinical data showed that 36.4% of patients developed peripheral neuropathy after treatment with linezolid. H&E staining showed that linezolid caused disordered arrangement of sciatic nerve tissue and increased space. The results of WB technique showed that linezolid-induced peripheral nerve toxicity was related to autophagy. CONCLUSION The mechanism of long-term linezolidin-induced peripheral neuropathy may be related to activation of glial autophagy.
