| 目的:比较麻黄汤配方颗粒与饮片汤剂解热效能的差异,为配方颗粒的临床使用提供一定的理论依据。方法: 将50只SD大鼠随机分为正常组、发热模型组、传统饮片麻黄汤剂组、配方颗粒麻黄汤剂组、布洛芬混悬液阳性对照组5组,每组各10只。除正常组外其余各组大鼠皮下注射20%酵母混悬液制备发热模型。造模后正常组、发热模型组大鼠均灌胃生理盐水2mL,其余各组大鼠灌胃相应的药液。于给药后不同时间点记录大鼠肛温,并使用ELISA试剂盒测定血浆中肿瘤坏死因子(TNF-α)、白细胞介素-1β(IL-1β) 、白细胞介素-6(IL-6)及下丘脑组织中前列腺素E2(PGE2)、环磷酸腺苷(cAMP)的含量。结果:肛温检测结果显示,与模型组相比,传统饮片麻黄汤剂组、配方颗粒麻黄汤剂组、布洛芬混悬液阳性对照组大鼠肛温都显著降低(P<0.05);与传统饮片麻黄汤剂组相比,配方颗粒麻黄汤剂组大鼠肛温无显著变化(P>0.05)。大鼠血浆ELISA检测结果显示,与模型组相比,传统饮片麻黄汤剂组和配方颗粒麻黄汤剂组大鼠血浆TNF-α、IL-1β和IL-6含量显著降低(P<0.05),布洛芬混悬液阳性对照组大鼠血浆TNF-α、IL-1β和IL-6含量显著降低(P<0.05或P<0.01);与传统饮片麻黄汤剂组比较,配方颗粒麻黄汤剂组和布洛芬混悬液阳性对照组血浆TNF-α、IL-1β和IL-6含量无显著性变化(P>0.05)。下丘脑组织ELISA检测结果显示,与模型组相比,传统饮片麻黄汤剂组大鼠cAMP含量显著降低(P<0.05),配方颗粒麻黄汤剂组和阳性对照组大鼠PGE2和cAMP含量显著降低(P<0.05或P<0.01);与传统饮片麻黄汤剂组相比,配方颗粒组大鼠cAMP含量显著降低(P<0.05或P<0.01)。结论:在相同药材剂量下,麻黄汤配方颗粒汤剂和麻黄汤传统饮片汤剂都有较强的解热效能。中药配方颗粒使用更方便,值得临床推广应用。 |

| Abstract Objective To compare differences between the antipyretic effect of Mahuang Tang formula granule and decoction , and to provide a theoretical basis for the clinical use of formula granule. Methods 50 SD rats were randomLy divided into normal group, fever model group, traditional Mahuang Tang decoction group , Mahuang Tang formula granule group and ibuprofen suspension positive control group, with 10 rats in each group.Fever models were established by subcutaneous injection of 20% yeast suspension into rats in all groups except the normal group.After modeling, normal group and fever model group were intragastrically given 2mL of physiological saline,and other groups were intragastrically given corresponding liquid medicine.The anal temperature of rats was recorded at different time points after administration, and the plasma levels of tumor necrosis factor (TNF-α), interleukin-1β(IL-1β), rleukin-6 (IL-6), prostaglandin E2(PGE2), cyclic adenosine monophosphate(cAMP) were determined by ELISA kit.Results The results of the anal temperature test show that, Compared with the model group, the anal temperature of rats in traditional Mahuang Tang decoction group, Mahuang Tang formula granule group and ibuprofen suspension positive control group were significantly decreased (P<0.05). Compared with traditional Mahuang Tang decoction group, the anal temperature of rats in the Mahuang Tang formula granule group had no significant change (P>0.05). The ELISA results of rat plasma showed that, Compared with the model group,the traditional Mahuang Tang decoction group and Mahuang Tang formula granule group rats plasma TNF-α, IL-1β, and IL-6 content is significantly decreased (P<0.05), and the ibuprofen suspension positive control group rat plasma TNF-α, IL-1β, and IL-6 is significantly decreased (P<0.05 or P<0.01); Compared with the traditional Mahuang Tang decoction group, Mahuang Tang formula granule group and ibuprofen suspension positive control group plasma TNF-α, IL-1β and IL-6 content have no significant changes (P> 0.05). The ELISA results of rat hypothalamic tissue showed that, Compared with the model group, the traditional Mahuang Tang decoction group rat cAMP content is significantly decreased (P<0.05), Mahuang Tang formula granule group and positive control group rats PGE2 and cAMP content is significantly decreased (P<0.05 or P<0.01); Compared with the traditional Mahuang Tang decoction group, Mahuang Tang formula granule group rat cAMP content is significantly decreased (P<0.05 or P<0.01). Conclusion Both the Mahuang Tang formula granule and traditional Mahuang Tang decoction have strong antipyretic effect under the same dose of medicinal materials. Traditional Chinese medicine formula granules are more convenient to use and are worthy of clinical promotion and application. |
