      目的 探讨厚胞汤调节成纤维细胞对防治宫腔粘连的作用机理。方法 将10只健康SD大鼠随机分成空白对照组和厚胞汤组,分别灌胃等量生理盐水、厚胞汤,2 mL/只·次,2 次/天以制备空白血清及含药血清,将经转化生长因子-β1(Transforming growth factor-β1,TGF-β1)刺激后的成纤维细胞分别加入空白血清及厚胞汤含药血清,分为空白对照组和厚胞汤组,采用CCK8法检测两组细胞增殖情况,流式细胞法检测细胞凋亡水平、细胞周期变化,Western blot检测两组细胞的TGF-β1、信号转导蛋白家族(Smads)表达水平。结果 与空白对照组比较,厚胞汤组的细胞增殖率显著降低(第2天, [(1.19±0.02)%比(1.05±0.04)%, P<0.01];第3天,[(1.36±0.02)%比(1.11±0.02)%, P<0.001];第4天,[(1.55±0.02)%比(1.18±0.02)%, P<0.001];第5天,[(1.89±0.03)%比(1.16±0.02)%, P<0.001]);细胞凋亡水平显著升高([(6.31±0.17)%比(10.19±0.72)%, P<0.01]);细胞周期G1期比例降低([(52.30±0.56)%比(48.78±1.51)%, P<0.05] ),G2期比例升高([(22.86±0.88)%比(27.56±0.64)%, P<0.01])。Western blot结果显示TGF-β1、Smad3表达显著下调。结论 厚胞汤可能通过TGF-β/ Smads通路有效抑制成纤维细胞纤维化表达,从而对宫腔粘连起到防治作用。

      Objective To investigate the mechanism of Houbao Recipe prescriotion and treatment of Intrauterine adhesions by regulating fibroblast. Methods In order to prepare the black serum and medicated serum,10 Healthy SD rats were randomly divided into control group and Houbao group,which gavaged the same amount of normal saline and Houbao Recipe(each 2 mL/time,twice/day).The fibroblast stimulated by TGF-β were divided into control group and Houbao group,which given black serum and medicated serum.The proliferation,apoptosis,cell cycle were analyzed by CCK8 assay, flow cytometry and western blotting. Result Compared with control group,the percentage of survival cells decreasing significantly(D2,[(1.19±0.02)% vs.(1.05±0.04)%, P<0.01];D3,[(1.36±0.02)% vs.(1.11±0.02)%, P<0.001];D4, [(1.55±0.02)% vs.(1.18±0.02)%, P<0.001];D5, [(1.89±0.03)% vs. (1.16±0.02)%, P<0.001]),as well as expression of TGF-β1、Smad3 treated by Houbao group, the apoptosis of Houbao group was significantly triggered([(6.31±0.17)% vs. (10.19±0.72)%, P<0.0001]).In the cell cycle assay, the percentage of Houbao group in G1 phase were significantly less than control group([(52.30±0.56)% vs. (48.78±1.51)%, P<0.05]),G2 phase were on the contrary([(22.86±0.88)% vs. (27.56±0.64)%, P<0.01]).Conclusion Houbao Recipe inhibit fibroblast expression by TGF-β/Smads probably to prescribe and treat Intrauterine adhesions.
