| 目的 探讨美洲大蠊研粉对脊髓半横断损伤(hSCI)大鼠脊髓损伤区域氧化应激的影响。方法 将60只雌性SD大鼠制作hSCI模型,用随机数字表法均分为对照组和观察组,对照组打开T10椎板行左侧脊髓半横断,术前30 min及术后每天灌胃生理盐水5 mL/只;观察组打开T10椎板行左侧脊髓半横断,术前30 min及术后每天灌胃美洲大蠊研粉生理盐水溶液(630 mg/kg)5 mL/只。术后14 d每组取5只大鼠用于尼氏染色;术后6 h及1、3、7、14 d每组每个时间点取5只大鼠,用酶联免疫吸附试验(ELISA)检测丙二醛(MDA)、8-羟基脱氧鸟苷(8-OhdG)的含量。结果 与对照组相比,观察组在10×20倍及10×40倍视野下脊髓神经元存活数目均更多[(26.25±0.50)个比(10.84±0.58),(14.92±0.32)个比(6.75±0.32)个,P<0.05];观察组术后7、14 d 脊髓组织MDA[(0.61±0.03)nmol/mL比(0.89±0.02)nmol/mL,(0.54±0.01)nmol/mL比(0.68±0.02)nmol/mL,P<0.05]和 8-OHdG[(17.92±0.99)ng/mL比(23.37±0.77)ng/mL,(15.91±0.28)ng/mL比(20.18±0.37)ng/mL,P<0.05]含量均低于对照组;与对照组相比,观察组氧化应激高峰时间更早,MDA和8-OHdG表达的峰值提前到了术后3d。结论 美洲大蠊研粉降低了hSCI大鼠脊髓损伤区域MDA、8-OHdG的含量,提前了氧化应激高峰时间,减轻了氧化应激损伤,减少了神经元丢失。 |

| Objective To investigate the effect of Periplanella American powder on oxidative stress in spinal cord injury area of rats with hemisection of spinal cord injury (hSCI).
Methods Sixty female SD rats were prepared as hSCI models, and they were divided into control group and observation group by random number table method, the control group opened the T10 vertebral plate and performed left spinal cord semi-transection, and each mouse was given 5ml of normal saline 30 min before operation and daily after surgery. The observation group opened the T10 lamina and performed semi-transection of the left spinal cord, and each mouse was given 5ml of normal saline solution of American cockroach grinding powder (630mg/kg) 30 min before operation and every day after operation.On the 14th day after operation, 5 mice were removed from each group for Nissl staining. At 6h, 1d, 3d, 7d, and 14 days after surgery, 5 mice were used in each group at each time point, and the contents of MDA (malondialdehyde) and 8-OhdG (8-hydroxydeoxyuridine) were detected by ELISA.
Results Compared with the control group, the number of spinal cord neurons survived in the 10×20x and 10×40x field of view in the observation group was higher[(26.25±0.50) vs.(10.84±0.58),(14.92±0.32) vs. (6.75±0.32),P<0.05]; The contents of MDA [(0.61±0.03)nmol/mL vs. (0.89±0.02)nmol/mL,(0.54±0.01)nmol/mL vs. (0.68±0.02)nmol/mL,P<0.05] and 8-OHdG [(17.92±0.99)ng/mL vs. (23.37±0.77)ng/mL,(15.91±0.28)ng/mL vs. (20.18±0.37)ng/mL,P<0.05] in spinal cord tissues in the observation group were lower than those in the control group at 7 and 14 days after surgery. Compared with the control group, the peak of oxidative stress in the observation group was earlier, and the peak expression of MDA and 8-OHdG was advanced to the third postoperative day.
Conclusion The Periplanella Americana powder reduced the content of MDA and 8-OHdG in the spinal cord injury area of hSCI rats, advanced the peak time of oxidative stress, alleviated oxidative stress damage, and reduced neuronal loss. |
