      目的:探究“补肾活血”针刺法对SAMP8小鼠认知功能和海马神经元SIRT2介导的RTN4B/BACE1通路的影响。方法:使用加速衰老的小鼠模型SAMP8(senescence-accelerated mouse-prone 8)。分别设正常对照组(SAMR1)6只、模型对照组(SAMP8)6只、针刺治疗组(SAMP8 + AP)6只、非穴位对照组(SAMP8 + CP)6只。针刺治疗组针刺选取百会、血海、肾俞、膈俞;血海、膈俞施捻转泻法,肾俞、百会施捻转补法,运针1 min,留针10 min。非穴位组则在非经非穴点进行刺激,干预 8周。通过Morris水迷宫(MWM)测试评估认知功能。采用免疫组织化学染色和ELASA评估海马神经元小胶质细胞的激活和Aβ沉积。通过WB和免疫组织化学检测SIRT2途径相关蛋白(SIRT2、RTN4B、BACE1和APP-CTF)的表达水平。结果:“补肾活血”针刺法改善了SAMP8小鼠的认知功能。“补肾活血”针刺法减少了海马组织中小胶质细胞介导的Aβ沉积,降低了SIRT2、BACE1和APP-CTF水平,并增加了RTN4B表达。结论:“补肾活血”针刺法通过抑制SIRT2介导的RTN4B/BACE1途径,减轻了SAMP8小鼠的认知缺陷和Aβ沉积。

      Objective: To investigate the effects of the "Bushen Huoxue" acupuncture on cognitive function and the SIRT2-mediated RTN4B/BACE1 pathway in hippocampal neurons of SAMP8 mice.. Methods: The senescence-accelerated mouse-prone 8 (SAMP8) model was utilized. Groups were established as follows: normal control group (SAMR1), model control group (SAMP8), acupuncture treatment group (SAMP8 + AP), non-acupoint control group (SAMP8 + CP), and drug treatment control group (SAMP8 + DPZ). Acupuncture treatment group selected the acupoints Baihui, Xuehai, Shenshu, and Geshu; the SAMP8 model control group and SAMR1 normal control group mice were subjected to the same duration and intensity of grasping stimulation as the other three groups, with an 8-week intervention period. Cognitive function was evaluated using the Morris Water Maze (MWM) test. Immunohistochemical staining was used to assess the activation of hippocampal neurons, microglia, and Aβ deposition. The expression levels of SIRT2 pathway-related proteins (SIRT2, RTN4B, BACE1, and APP-CTF) were examined by Western Blot (WB). Results: Bushen Huoxue Acupunctureimproved cognitive function in SAMP8 mice. It reduced microglial-mediated Aβ deposition in the hippocampus, decreased levels of SIRT2, BACE1, and APP-CTF, and increased RTN4B expression. Conclusion: Bushen Huoxue Acupuncture alleviated cognitive deficits and Aβ deposition in SAMP8 mice by inhibiting the SIRT2-mediated RTN4B/BACE1 pathway.
