      目的? 探讨配对相关同源框1(PRRX1)、基质金属蛋白酶13(MMP13)在胃癌(GC)组织中的表达及其与临床病理特征和预后的关系。方法 利用Ualcan数据库检索GC/正常胃组织中PRRX1 mRNA、MMP13 mRNA的表达水平,并选取差异明显的PRRX1及MMP13作为研究靶点。采用实时荧光定量PCR检测GC组织/癌旁组织PRRX1 mRNA、MMP13 mRNA表达水平、探讨其表达水平与临床病理特征、预后的关系。采用免疫组织化学法检测PRRX1、MMP13蛋白表达。结果? Ualcan数据库显示,GC组织PRRX1 mRNA、MMP13 mRNA表达水平高于正常胃组织(P<0.05);GC组织中PRRX1 mRNA、MMP13 mRNA表达水平均高于癌旁组织(P<0.05);GC组织PRRX1 mRNA、MMP13 mRNA表达水平与分化程度、脉管癌栓、淋巴结转移、浸润程度和TNM分期相关(P<0.05);GC组织中PRRX1 mRNA表达水平与MMP13 mRNA呈正相关(P<0.05);PRRX1 mRNA高表达组、MMP13 mRNA高表达组累积生存率均低于PRRX1 mRNA低表达组、MMP13 mRNA(P<0.05)。免疫组织化学分析显示,与癌旁正常组织相比,肺癌组织中RRX1、MMP13蛋白高表达率明显升高(P<0.05)。结论? GC组织中PRRX1 mRNA、MMP13 mRNA表达水平升高,二者均与GC临床病理特征和预后有关,可为临床诊治GC提供一定参考。

      Objective To investigate the expression of paired related homeobox 1 (PRRX1) and matrix metalloproteinase 13 (MMP13) in gastric cancer (GC) tissues and its relationship with clinicopathological features and prognosis. Methods The expression levels of PRRX1 mRNA and MMP13 mRNA in GC/normal gastric tissues were retrieved using the Ualcan database, and PRRX1 and MMP13, which had significant differences, were selected as the targets of the study. The expression levels of PRRX1 mRNA and MMP13 mRNA in GC tissues/paracancerous tissues were detected by real-time fluorescence quantitative PCR, and the relationship between the expression levels with clinicopathological features and prognosis were investigated. PRRX1, MMP13 protein expression were detected by immunohistochemistry. Results Ualcan database showed that the expression levels of PRRX1 mRNA and MMP13 mRNA in GC tissues were higher than those in normal gastric tissues (P<0.05); the expression levels of PRRX1 mRNA and MMP13 mRNA in GC tissues were higher than those in adjacent tissues (P<0.05); the expression levels of PRRX1 mRNA and MMP13 mRNA in GC tissues were correlated with the degree of differentiation, vascular cancer embolism, lymph node metastasis, degree of infiltration, and TNM stage (P<0.05); the expression of PRRX1 mRNA in GC tissues was positively correlated with MMP13 mRNA (P<0.05); the cumulative survival rate of PRRX1 mRNA overexpression group and MMP13 mRNA overexpression group was lower than that of PRRX1 mRNA lowexpression group and MMP13 mRNA lowexpression group (P<0.05). Immunohistochemical analysis showed that the high expression rate of RRX1 and MMP13 proteins was significantly higher in lung cancer tissues compared with normal tissues adjacent to the cancer (P < 0.05). Conclusion The expression levels of PRRX1 mRNA and MMP13 mRNA were elevated in GC tissues, both of which were related to the clinicopathological features and prognosis of GC, and could provide some reference for the clinical diagnosis and treatment of GC.
