| 【目的】探讨黄连素对幽门螺杆菌感染小鼠胃内环境的影响。【方法】选取5-6周BALB/C雄性SPF级小鼠,适应性饲养1周后,随机分为四组,分别为正常组、正常+黄连素组、模型组以及模型+黄连素组。小鼠禁食12小时后,以HpSS1菌液灌胃,隔天1次,持续3天。鉴定造模成功后,对正常+黄连素组和模型+黄连素组进行黄连素灌胃治疗,连续7天。4周后对所有小鼠进行处死取材。酸碱滴定法测定进行胃酸、ELISA法进行胃蛋白酶原、白介素-6(interleukin-6, IL-6)、白介素-8(IL-8)、白介素-10(IL-10)测定及HE染色观察各组胃粘膜病理改变。【结果】与正常组pH值(5.19±1.99)相比,模型组的胃液pH值(2.67±0.78)呈显著下降,而模型+黄连素组胃液的pH值(4.83±1.03)显著上升。模型组中胃蛋白酶原水平5.18±1.19pg /mL),较正常组(2.01±1.11pg /mL)及正常+黄连素组(2.37±1.04pg /mL)相比,呈显著升高,而模型+黄连素组中胃蛋白酶原水平(3.74±0.91pg /mL)有下降,差异具有统计学意义(p<0.05);模型组中IL-6(15.12±4.37pg /mL)、IL-8(42.36±5.12pg /mL)、IL-10(16.32± 5.34pg /mL)水平较正常组IL-6(5.19±1.99pg /mL)、IL-8(22. 59±2.34pg /mL)、、IL-10(4. 59±1. 24pg /mL)及正常+黄连素组IL-6(4.79±2.11pg /mL)、IL-8(20.45±3.37pg /mL)、IL-10(4.06±1.44pg /mL)显著升高;而模型+黄连素组IL-6(7.27±1.53pg /mL)、IL-8(27.56±4.31pg /mL)、IL-10(8.32±1.78pg /mL)水平呈显著下调。病理结果可以发现,正常组和正常+黄连素组镜下观察可见胃粘膜及粘膜下层结构清晰,腺体完整,未见病理改变,模型组镜下观察可见,胃粘膜结构破坏,可见大量炎细胞浸润,而模型+黄连素组观察发现胃粘膜结构尚完整,只见少量炎细胞浸润。【结论】黄连素可缓解幽门螺杆菌感染小鼠胃粘膜炎症反应,改善胃内环境。 |

| Objective To investigate the effects of berberine on the gastric environment in mice infected with Helicobacter pylori.Methods 5-6 week-old BALB/C male SPF mice were acclimated for 1 week and randomly divided into four groups: the normal group, the normal+berberine group, the HP infection group, and the HP infection+berberine group. After fasting for several hours, the mice were administered with HpSS1 bacterial suspension via gastric lavage, once a day for 3 consecutive days. After confirming the establishment of the model, the normal+berberine group and the model+berberine group were given continuous gastric lavage with berberine for 7 days. All mice were sacrificed and tissues were collected 4 weeks later.The gastric acid was determined by acid-base titration, and the gastric pepsinogen, interleukin-6 (IL-6),interleukin-8 (IL-8), and interleukin-10 (IL-10) were measured by ELISA. The pathological changes in gastric mucosa of each group were observed by HE staining.Results Compared with the normal group (5.19±1.99), the pH value of gastric juice in the group (2.67±0.78) decreased significantly, while the pH value of gastric juice in the model berberine group (483±1.03) increased significantly. The level of pepsinogen in the model group (5.18±1.19 pg) was significantly higher than that in the normal group (2.01±1.11 pg/mL) and the normal berberine group (.37±1.04 pg/mL), while the level of pepsinogen in the model berberine group (3.740.91 pg/mL) decreased, with a significant difference (p<0.05). The levels of IL-6 (15.1±4.37 pg/mL), IL-8 (42.36±5.12 pg/mL), and IL-10 (1632±5.34 pg/mL) in the model group were significantly higher than those in the normal group (IL-6: 5.1±1.99 pg/mL, IL-8: 22.59±2.34 pg/mL, IL-10: 459±1.24 pg/mL) and the normal berberine group (IL-6: 4.79±2.1 pg/mL, IL-8: 20.45±3.37 pg/mL, IL-10: 4.06±144 pg/mL). However, the levels of IL-6 (7.27±1.53 pg/mL), IL-8 (2756±4.31 pg/mL), and IL-10 (8.32±1.78 pg/mL) in the model berine group were significantly lower. Pathological results showed that the gastric mucosa and submucosa in the normal group and the normal berberine group clearly visible, with intact glands and no pathological changes. In the model group, the gastric mucosa was destroyed, with a large number of inflammatory cells infiltrating In the model berberine group, the gastric mucosa was intact, with only a few inflammatory cells infiltrating.Conclusion The study shows that berberine can alleviate the gastric mucosal inflammation caused by Helicobacter pylori in mice, thereby improving the internal environment and playing a role in the treatment of helicobacter pylori associated gastritis to a certain extent. |
