| 目的 探讨核孔蛋白Nup93复合体基因对肝癌预后的预测价值及其潜在的分子机制。方法 对癌症基因组图谱(TCGA)和国际癌症基因组联盟(ICGC)数据库的共574例肝癌患者进行回顾性RNA-seq数据分析。比较肝癌与正常肝组织的基因差异表达,通过ROC曲线与Logistic回归分析基因对肝癌的诊断效能,采用Kaplan-Meier法对核孔蛋白Nup93复合体基因高低表达组的生存期进行比较。通过基因功能富集、基因集变异分析以及Pearson相关分析等方法来探索核孔蛋白Nup93复合体基因与细胞周期过程、细胞周期分子以及信号通路因子的联系。基于单细胞测序数据集,探索核孔蛋白Nup93复合体基因在肿瘤微环境中的表达模式。结果 核孔蛋白Nup93复合体基因在肝癌中高表达,并与肝癌患者不良预后密切相关。核孔蛋白Nup93复合体基因参与肝癌细胞周期调控,与有丝分裂周期以及肿瘤增殖过程有关,并在增殖性T细胞中富集。结论 核孔蛋白Nup93复合体基因可作为肝癌的潜在生物标志物和肝癌患者总生存期的预测因子,涉及肝癌细胞周期调控。核孔蛋白Nup93复合体基因有望成为肝癌治疗策略的新靶点。 |

| Objective To investigate the predictive value of Nup93 complex gene in hepatocellular carcinoma and its potential molecular mechanism. Methods The RNA-seq data of 574 liver cancer patients from the Cancer Genome Atlas (TCGA) and the International Cancer Genome Consortium (ICGC) were retrospectively analyzed. The differential expression of genes in liver cancer and normal liver tissues was compared. ROC curve and Logistic regression were used to analyze the diagnostic efficiency of genes for liver cancer. Kaplan-Meier method was used to compare the survival of the group with high or low expression of Nup93 complex gene. Gene function enrichment, gene set variation analysis and Pearson correlation analysis were used to explore the association of Nup93 complex gene with cell cycle processes, cell cycle molecules and signaling pathway factors. To explore the expression pattern of Nup93 complex gene in tumor microenvironment based on single-cell sequencing dataset. Results The nucleoporin Nup93 complex gene was highly expressed in hepatocellular carcinoma and closely linked to a poor prognosis for patients with hepatocellular carcinoma. The nucleoporin Nup93 complex gene is involved in the regulation of HCC cell cycle, which is related to mitotic cycle and tumor proliferation, and is enriched in proliferative T cells. Conclusion Nup93 complex gene can be used as a potential biomarker of hepatocellular carcinoma and a predictor of overall survival of patients with hepatocellular carcinoma, which is involved in cell cycle regulation. The nucleoporin Nup93 complex gene is expected to be a new target for liver cancer treatment strategies. |
