| 目的 探究中医药治疗慢性前列腺炎湿热瘀滞证(Chronic Prostatitis of Dampness-heat and Blood-stagnancy Type,CP-DHBS)的组方规律,预测核心药物组合并挖掘相关作用机制,为后续实验研究和疾病防治提供指导和依据。方法 检索中英文数据库从建库至2024年6月关于中药治疗CP-DHBS的文献,筛选符合纳入标准的文献,构建慢性前列腺炎湿热瘀滞证处方数据库。使用数据挖掘中关联规则和聚类分析等算法研究中医组方规律;使用网络药理学方法研究核心药物组合的成分、靶点,分析其涉及的GO功能和KEGG通路富集,从而阐明其作用机制。结果 本研究共纳入62篇文献、64首处方,涉及中药162味,这些药多味苦、甘,性寒,归肝经。关联规则分析结果显示强关联四药组合“桃仁-红花-败酱草-丹参”是核心药物组合。“桃仁-红花-败酱草-丹参”治疗CP-DHBS关键成分包括:槲皮素、木犀草素、山奈酚等;关键靶点包括:TP53、AKT1、STAT3、IL6、TNF等;主要生物过程为:基因表达的正向调控、凋亡过程负向调控、RNA聚合酶Ⅱ转录的正向调控等等;涉及的信号通路主要有癌症通路、前列腺癌、TNF信号通路等。结论 中医药治疗CP-DHBS组方用药遵循清热利湿,活血化瘀的原则,辅以解毒、止痛的同时,注重气血和调;核心药物组合“桃仁-红花-败酱草-丹参”的作用机制主要与炎症反应、免疫调控等生物学过程相关,为中药治疗CP-DHBS提供理论依据,为发掘其潜在作用机制提供方向。 |

| Objective To explore the formula formulation rule of Chronic Prostatitis of Dampness-heat and Blood-stagnancy Type (CP-DHBS) in the treatment of chronic prostatitis, predict the core drug combination and explore the relevant mechanism. To provide guidance and basis for follow-up experimental research and disease prevention. Methods The literature on CP-DHBS with Chinese medicine was searched from the establishment of Chinese and English databases to June 2024, and the literature meeting the inclusion criteria was screened to construct the prescription database of damp-heat stasis syndrome of chronic prostatitis. The association rules and cluster analysis algorithms in data mining were used to study the rules of TCM prescription formation. The components and targets of the core drug combination were studied by network pharmacology, and the GO function and KEGG pathway enrichment were analyzed to elucidate the mechanism of action. Results A total of 62 literatures and 64 prescriptions were included in this study, involving 162 traditional Chinese medicines, which were bitter, sweet, cold and classified into the liver channel. The results of association rule analysis showed that the strong association four-drug combination "peach kernel, safflower, patrinia and salvia miltiorrhiza" was the core drug combination. The key ingredients of "peach kernel, safflower, patrinia and salvia miltiorrhiza" treatment for CP-DHBS include quercetin, luteolin, kaempferol, etc. Key targets include TP53, AKT1, STAT3, IL6, TNF, etc. The main biological processes are: positive regulation of gene expression, negative regulation of apoptosis, positive regulation of RNA polymerase II transcription, etc. The signaling pathways involved mainly include cancer pathway, prostate cancer pathway, TNF signaling pathway, etc. Conclusion The treatment of CP-DHBS with traditional Chinese medicine should follow the principle of clearing heat and dampness, promoting blood circulation and removing blood stasis, and pay attention to Qi and blood regulation while supplementing with detoxification and pain relief. The mechanism of action of the core drug combination "peach kernel - Safflower - Patrinia - Salvia Miltiorrhiza" is mainly related to biological processes such as inflammatory response and immune regulation, which provides a theoretical basis for the treatment of CP-DHBS with traditional Chinese medicine and provides a direction for exploring its potential mechanism of action. |
