| 目的 本研究旨在探讨白藜芦醇(Res)对小鼠腹主动脉瘤(AAA)形成的抑制作用及其潜在机制。方法 实验采用10周龄雄性C57/B6小鼠,随机分配至正常组、模型组、低剂量Res组(30 mg/kg/d)及高剂量Res组(120 mg/kg/d)。模型组通过腹腔注射苯并芘(BaP)并结合血管紧张素Ⅱ(AngⅡ)给药以诱导AAA的形成。给药组则分别予不同剂量Res灌胃。经过四周处理后,收集小鼠血清样本检测肿瘤坏死因子-ɑ(TNF-ɑ)水平,同时行生化检测,评估血脂情况;取靶血管制作石蜡切片,并进行HE染色观察大动脉的结构变化,同时测量腹主动脉的平均周长并测定AAA形成率。免疫组化法评估各组动脉壁基质金属蛋白酶-9(MMP-9)的阳性情况。结果 1、 形态学结果显示,正常组腹主动脉平均周长(912±141)m,模型组周长(1271±118)m,与高剂量Res组周长(1083±120)m比较差异有统计学意义,p <0.05。2、血标本检测显示,Res显著降低各组小鼠TNF-α及血脂水平。3、各组动脉壁MMP-9比较,模型组(0.51±0.11)较正常组(0.17±0.08)显著升高;高剂量Res组(0.30±0.08)较模型组存在下降趋势, p <0.05。结论 Res抑制炎症反应,降脂延缓动脉硬化同时减少血管壁基质降解,维持小鼠动脉壁结构完整性以抑制AAA形成从而发挥保护作用 |

| ABSTRACT Objective To investigate the inhibitory effect of resveratrol (Res) on the formation of abdominal a aneurysm (AAA) in mice and explore its related mechanisms. Methods Ten-week-old male C57/B6 mice were randomly divided into normal group,model group, low-dose Res group (30 mg/kg/d), and high-dose Res
group (120 mg/kg/d). The model group was BaP intraperitoneal injection combined
with angiotensin II administration to construct the AAA model, and the administration
group was also given Res gavage at the dose. After 4 weeks, mouse serum biochemical tests were performed to detect lipid content, and target vessels were taken for paraffin sectioning and HE staining to the structural changes of the aorta, and the average vascular circumference of the abdominal aorta and the formation rate of abdominal aortic aneurysm were measured. positive expression of matrix metalloproteinase 9 (MMP-9) in the vascular wall was detected by immunohistochemistry. Results l.logical results showed that the average circumference of the abdominal aorta in the normal group was (912±141)m, the circumference in the group was (1271±118) m, and the difference was statistically significant compared with the high-dose Res group (1083±120)m, p < 0.05. 2. Blood sample tests suggested that Res reduced TNF-ɑ and lipid content in mice in all groups. 3. Immunohistochemical results that compared with the normal group MMP9 (0.17±0.08), the model group (0.51±0.11) was significantly increased, and the high-dose Res group (0.30±0.08) was decreased to a certain extent, p <0.05. Conclusion Res can inhibit the formation of abdominal aortic aneurysm, and its mechanism may be to inhibit the inflammatory response, reduce content to delay atherosclerosis, and reduce the degradation of vascular wall matrix. Maintaining the structural integrity of the artery wall in mice inhibited the formation of abdominal aortic aneurysm and played a protective role. |
