| 目的 探究南葶苈子水提物拮抗结直肠癌生长作用及潜在机制。方法 24只4周龄C57BL/6小鼠购于上海斯莱克实验动物有限责任公司,应用皮下注射SW620细胞构建结直肠癌异种移植小鼠模型,分为模型组、南葶苈子低、中、高剂量组,记录实验期间小鼠瘤体积和瘤重。流式细胞术检测肿瘤活性氧(reactive oxygen species, ROS)表达;免疫组化检测肿瘤增殖标志物Ki67和蛋白质二硫键异构酶(protein disulfide isomerase, PDI/PDIA1)水平;Western blot检测PDIA1蛋白表达。结果 与模型组相比,南葶苈子中、高剂量组小鼠瘤重[(0.56±0.13) g、(0.58±0.16) g比(1.46±0.24) g,P<0.01]和第9、12、15天的肿瘤体积[(397.86±59.11) mm3、(408.47±75.18) mm3比(610.13±76.02) mm3,P<0.01]、[(489.89±79.73) mm3、(582.96±172.19) mm3比(1098.3±102.09) mm3,P<0.01]、[(625.46±171.17) mm3、(675.36±201.67) mm3比(1484.60±242.64) mm3,P<0.01]均显著降低,肿瘤组织细胞中ROS水平[(22.53±2.00)%、(28.54±1.91)%比(7.42±1.31)%,P<0.01]显著升高,肿瘤组织中Ki67[(37.68±4.60)、(29.47±5.09)比(60.45±5.66),P<0.01]、PDIA1[(13.33±1.49)、(9.75±0.57)比(20.23±1.29),P<0.01]均显著下降,PDIA1蛋白表达水平[(0.310±0.065)、(0.178±0.060)比(1.000±0.119),P<0.01]显著降低。结论 南葶苈子水提物能显著抑制结直肠癌生长,其机制可能与促进ROS表达、抑制PDIA1作用有关。 |

| To explore the effect and potential mechanism of the total extract of Descurainiae semen on the growth of colorectal cancer. Methods Twenty-four 4-week-old C57BL/6 mice were purchased from Shanghai Slac Laboratory Animal Co., Ltd. The xenotransplantation mouse model of colorectal cancer was established by subcutaneous injection of SW620 cells. The mice were divided into model group, low, medium and high dose groups of Descurainiae semen. The tumor volume and tumor weight were recorded during the experiment. The expression of ROS was detected by flow cytometry. The levels of tumor markers Ki67 and protein disulfide isomerase (PDIA1) were detected by immunohistochemistry. The expression of PDIA1 protein was detected by Western blot. Results Compared with the model group, the medium- and high-dose groups of Semen Lepidii showed significantly reduced tumor weight [(0.56±0.13) g and (0.58±0.16) g vs. (1.46±0.24) g, P<0.01] and tumor volume on days 9, 12, and 15 [(397.86±59.11) mm3, (408.47±75.18) mm3 vs. (610.13±76.02) mm3, P<0.01]; [(489.89±79.73) mm3, (582.96±172.19) mm3 vs. (1098.3±102.09) mm3, P<0.01]; [(625.46±171.17) mm3, (675.36±201.67) mm3 vs. (1484.60±242.64) mm3, P<0.01], respectively. Additionally, the ROS levels in tumor tissue cells were significantly increased [(22.53±2.00)%, (28.54±1.91)% vs. (7.42±1.31)%, P<0.01], while the expression of Ki67 [(37.68±4.60), (29.47±5.09) vs. (60.45±5.66), P<0.01] and PDIA1 [(13.33±1.49), (9.75±0.57) vs. (20.23±1.29), P<0.01] in tumor tissues was significantly decreased. The protein expression level of PDIA1 was also significantly reduced [(0.310±0.065), (0.178±0.060) vs. (1.000±0.119), P<0.01]. Conclusion The total extract of Descurainiae semen decoction can significantly inhibit the growth of colorectal cancer, and its mechanism may be related to promoting ROS expression and inhibiting PDIA1 action. |
