      Objective To establish a transdermal drug delivery evaluation system based on the pathological characteristics of eczema skin and promote the development of novel external preparations for clinical empirical formula No. 1 for Eczema. Methods Establish a quality control system and employ transdermal penetration technology to investigate cumulative penetration amount and skin retention amount, and study the effects of various dosage forms on skin structure, in order to screen out the optimal modern topical dosage form for the clinical empirical formula No.1 for Eczema. Results The gel formulation of clinical empirical formula No. 1 for Eczema demonstrated superior retention capacity of its active ingredients in both the epidermal and dermal layers,higher cumulative permeation amount per unit area, and enhanced ability to penetrate the stratum corneum.The gel formulation of the Clinical Experience Formula No.1 for eczema exhibits enhanced retention capacity with higher cumulative drug permeation per unit area, demonstrating superior ability to penetrate the stratum corneum. Conclusion This study innovatively integrates a transdermal drug delivery evaluation system, establishing a standardized technical pathway for the translational research of classical renowned prescriptions into topical formulations.
