| [目的] 基于数据挖掘和网络药理学分析中医肺病专家肺结节用药规律及作用机制。[方法] 收集葛琳仪工作室中医肺病专家治疗肺结节的病案处方,对中药处方药物进行聚类分析、关联规则等数据挖掘。检索相关数据库获得核心组方的活性成分、作用靶点及疾病的作用靶点,并进行韦恩分析得到药物与疾病的交集靶点。进一步绘制“药物-化合物-靶点基因”网络图以及蛋白质互作网络图。分析上述交集靶点基因的富集功能和通路并进行可视化。[结果] 中药处方中四性五味以温、平、苦、辛为主,归经为肝、脾、肺为主,功效以活血化瘀、补虚为主。二项强关联规则分析发现,“三棱 => 莪术”,“红花 => 桃仁”为主要的二项规则。核心组方的有效活性化合物为槲皮素、木犀草素、山奈酚等,关键靶点为白细胞介素-1B、肿瘤蛋白53、环氧合酶2等。生物学过程主要富集于基因表达的正向调控、凋亡过程负向调控等;细胞定位主要富集于细胞外间隙、细胞外区等;分子功能主要富集于酶结合、相同蛋白结合等。核心组方治疗肺结节主要是通过癌症信号通路等。[结论] 益气健脾、行气活血、化痰散结为肺结节治疗基本方法,核心药物通过多成分、多靶点、多通路发挥作用,可为肺结节的临床治疗和机制研究提供参考。 |

| ?[?Objective]? To analyze the medication patterns and mechanisms of core herbal formulas for pulmonary nodules by integrating data mining and network pharmacology approaches, based on clinical prescriptions from traditional Chinese medicine (TCM) experts in the Ge Linyi Studio. [?Methods]? Prescriptions from TCM experts specializing in pulmonary diseases were collected and analyzed through cluster analysis, association rules? and association rule mining. Active components and therapeutic targets of the core herbal formula, along with disease-related targets, were retrieved from relevant databases. Venn analysis was performed to identify intersection targets between the herbal formula and the disease. A "TCM - compound - target" network and a protein-protein interaction (PPI) network were subsequently constructed. GO function enrichment analysis and KEGG path enrichment analysis were performed for intersection targets. [?Results]? The herbal properties were predominantly warm, neutral, bitter, and pungent, with primary meridian tropism to the liver, spleen, and lung meridians. The main therapeutic effects included activating blood circulation, resolving stasis, and tonifying deficiency. The binomial strong association rule analysis found that "san leng-e zhu", "hong hua-tao ren". The core formula contained active compounds such as quercetin, luteolin, kaempferol, targeting critical genes like IL1B, TP53, PTGS2. Biological process was mainly enriched in positive regulation of gene expression, and negative regulation of apoptotic process, and so on. Cellular component was mainly enriched in extracellular space, extracellar region, and so on. Molecular function was mainly enriched in enzyme binding, identical protein binding, and so on. The core prescription for treatment of pulmonary nodules was mainly through Pathways in cancer, and so on. [?Conclusion]? The fundamental TCM strategy for pulmonary nodules emphasizes reinforcing Qi, invigorating the spleen, promoting blood circulation, and resolving phlegm-stasis. The core formula operates through multi-component, multi-target, multi-pathway mechanisms, providing a scientific basis for clinical management and mechanistic research. |
