      【】目的:前期研究证实SIRT6在甲状腺乳头状癌(PTC)中表达上调且会影响PTC的侵袭及迁移,本研究意在探索SIRT6是否通过HIF-1α对PTC的EMT及恶性生物学行为的影响。方法:构建SIRT6敲低细胞系,后采用Western blotting及 RT-qPCR检测EMT相关标志物(E-cadherin, Vimentin, Snail)、HIF-1α及生物学行为实验(侵袭及迁移实验)。抑制HIF-1α后采用划痕实验探索PTC细胞恶性生物学行为变化。结果:敲低SIRT6后,K1及TPC-1细胞体外迁移及侵袭能力均有所下降。敲低SIRT6后的PTC细胞HIF-1α表达下降,与EMT相关的标志物,β-catenin,N-cadherin表达下调而E-cadherin表达上调。实验采用YC-1选择性抑制了HIF-1α的表达,结果表明SIRT6敲低细胞的迁移能力减弱,从而说明SIRT6/HIF-1α影响了PTC细胞的迁移能力。结论: SIRT6可以调控EMT及HIF-1α的表达。SIRT6/HIF-1α可以通过EMT影响甲状腺乳头状癌细胞的迁移及侵袭能力。

      【】Objective: Previous studies have confirmed that SIRT6 is up-regulated in papillary thyroid carcinoma (PTC) and affects the invasion and migration of PTC. This study is to explore whether SIRT6/ HIF-1α axis influences the EMT and malignant biological behavior of PTC. Methods: The SIRT6 knockdown cell line was constructed. In the SIRT6 knockdown cell line, the expression of EMT-related markers (E-cadherin, Snail) and HIF-1α was detected by Western blotting and RT-qPCR, and biological behavior experiments (invasion and migration experiments) were also performed. After inhibiting HIF-1α, wound healing assays was used to explore the change of malignant biological behavior of PTC cells. Results: After knocking down SIRT6, the migration and invasion ability of K1 and TPC-1 cells decreased in vitro. The expression of HIF-1α in SIRT6-knockdown PTC cells was decreased. For the markers associated with EMT, β-catenin and N-cadherin expression were down-regulated and E-cadherin expression was up-regulated. The expression of HIF-1α was selectively inhibited by YC-1. The results showed that the migration ability of SIRT6-knockdown PTC cells decreased in wound healing assays. Conclusion: SIRT6 regulates the expression of EMT and HIF-1α. SIRT6/HIF-1α can affect the migration and invasion of PTC cells through EMT.
