| 目的 探究细梗香草总皂苷(LC)协同顺铂抗肺癌效果及对机体血清免疫因子的调节机制。方法 BALB/c-nu/nu小鼠皮下接种A549细胞,构建肺癌移植瘤模型,将其随机分为对照(NC)组、LC低剂量组、LC中剂量组、LC高剂量组、顺铂(DDP)组、LC中剂量+顺铂联合治疗组。检测记录各组小鼠给药前后生长状态、体质量改变、移植瘤体积和脾指数等,免疫印迹(WB)检测移植瘤组织B细胞淋巴瘤-2相关X蛋白(Bax)、B细胞淋巴瘤-2蛋白(Bcl-2)、核因子κB(NF-κB)表达情况,酶联免疫吸附(ELISA)法检测血清白介素-2(IL-2)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)和干扰素-γ(IFN-γ)水平。结果 与NC组相比,各给药组(LC低、LC中、LC高、DDP和联合治疗组)小鼠体质量(减轻百分率分别为1.97%、6.34%、11.71%、24.95%和15.6%)和移植瘤体积(抑瘤率分别为12.77%、23.63%、33.33%、40.44%和64.21%)均有所下降,移植瘤组织Bcl-2、NF-κB蛋白和血清IL-6水平降低,Bax蛋白、IL-2、TNF-α和IFN-γ水平升高。与DDP组相比,联合治疗组移植瘤体质量[(0.524±0.087)g比(0.872±0.129)g,P<0.05]和IL-6水平[(23.82±3.57)pg/ml比(32.25±2.22)pg/ml,P<0.05]显著减小,脾脏指数[(5.53±0.30)mg/g比(3.11±0.53)mg/g,P<0.05]和IFN-γ水平[(454.64±65.73)pg/ml比(331.21±29.14)pg/ml,P<0.05]显著升高,且联合治疗组Bcl-2和NF-κB蛋白表达水平较其他各给药组变化更显著(P<0,05),合并用药疗效q>1。结论 细梗香草总皂苷可协同增效顺铂对肺癌的治疗效果,减轻化疗引起的不良反应,其作用机制可能与LC调节免疫因子和凋亡相关蛋白的表达有关。 |

| Objective To investigate the auxiliary therapeutic effect of Lysimachia capilliposide (LC) combined with cisplatin on lung cancer and its regulatory effect on immune factors. Methods The lung cancer model was established by hypodermic injection with the A549 cells in BALB/c-nu/nu mice. The model mice were randomly divided into 6 groups: control group, low-dose LC group, medium-dose LC group, high-dose LC group, cisplatin (DDP) group, and medium-dose LC + cisplatin combination group. After drug administration, the growth status, body weight changes, xenograft tumor volume, tumor inhibition rate, and spleen index of nude mice in each group were detected. Western blotting (WB) was used to detect the expression of B-cell lymphoma-2 (Bcl-2), bcl-2-associated X protein (Bax), and nuclear factor kappa-B (NF-κB) in xenograft tumor tissues, and enzyme-linked immunosorbent assay (ELISA) was used to detect the serum levels of interleukin-2 (IL-2), interleukin-6 (IL-6), tumor necrosis factor-α(TNF-α), and interferon-γ (IFN-γ). Results Compared with the control group, the body weight (Percent weight loss: 1.97%, 6.34%, 11.71%, 24.95%, and 15.6%) and xenograft tumor volume (Inhibition rate: 12.77%, 23.63%, 33.33%, 40.44%, and 64.21%) of mice in low-dose LC, medium-dose LC, high-dose LC, DDP and combination group decreased; the levels of Bcl-2 and NF-κB proteins in xenograft tumor tissues and serum IL-6 decreased, while the levels of Bax protein, IL-2, TNF-α, and IFN-γ increased. Compared with the DDP group, the xenograft tumor weight [(0.524±0.087) g vs. (0.872±0.129) g, P<0.05] and IL-6 level [(23.82±3.57) pg/ml vs. (32.25±2.22) pg/ml, P<0.05] in the combination therapy group were significantly reduced, and the spleen index [(5.53±0.30) mg/g vs. (3.11±0.53) mg/g, P<0.05] and IFN-γ level [(454.64±65.73) pg/ml vs. (331.21±29.14) pg/ml, P<0.05] were significantly increased. In addition, the changes in the expression of Bax, Bcl-2, and NF-κB proteins in the combination therapy group were more significant than those in other drug-administered groups (P<0.05), and the therapeutic efficacy index (q value) of combined drug use was greater than 1 (q>1). Conclusions Lysimachia capilliposide can assist in enhancing the therapeutic effect of cisplatin on lung cancer, improve the body''s immunity, and reduce adverse reactions caused by chemotherapy. Its mechanism may be related to the regulation of the expression of immune factors and apoptosis-related proteins by LC. |
