      【】 目的 研究黄芪甲苷对H2O2诱导的PC12细胞凋亡的保护作用及机制。方法 使用不同浓度黄芪甲苷干预经H2O2诱导的PC12细胞,ROS试剂盒检测PC12细胞内ROS含量,凋亡试剂盒检测PC12细胞凋亡率,Western blot检测PC12细胞BCL-2,Caspase-3,Bax等凋亡指标及PI3K/AKT信号通路关键蛋白表达。结果 0.8μmol/L黄芪甲苷可进一步提高PC12细胞存活率;中高黄芪甲苷剂量组PC12细胞内ROS及凋亡率显著下降;中高黄芪甲苷剂量组通过活化PI3K/AKT信号通路减低PC12细胞BCL-2,Caspase-3,Bax等凋亡指标蛋白水平。结论 中高剂量黄芪甲苷可基于PI3K/AKT信号通路改善经H2O2诱导的PC12细胞氧化应激及凋亡水平。

      【】 Objective To explore the protective effect of astragaloside on H2O2-induced apoptosis of PC12 cells and its mechanism. Methods Different concentrations of astragaloside were used to intervene PC12 cells induced by H2O2. The ROS content in PC12 cells was detected by ROS kit, t Apoptotic indicators such as BCL-2, Caspase-3 and Bax and the expression of key proteins in PI3K/AKT signaling pathway were detected by Western blot. Results 0.8μmol/L astragaloside could further improve the survival rate of PC12 cells. The ROS and apoptosis rates of PC12 cells were significantly decreased in the medium-high astragaloside dose group. The levels of apoptosis index proteins such as BCL-2, Caspase-3 and Bax in PC12 cells were decreased by activating PI3K/AKT signaling pathway in the medium-high astragaloside dose group. Conclusion Medium-high dose astragaloside can improve the oxidative stress and apoptosis of PC12 cells induced by H2O2 based on PI3K/AKT signaling pathway.
